Ⅳ。结语
基因治疗要成为可以广泛应用的医疗技术,在基因导入法方面有两个问题需要解决
。第一,要建立能稳定转导细胞的体内方法,最好还能有组织、器官特异性。第二
,基因进入目标细胞后,要能够安全地整合到染色体的非关键部位,或者恰与所要
修复的缺陷基因同源重组,总之必须防止诱发恶变。
病毒感染途径的导入法主要优点在于基因转移和表达的效能高,脂质体等非病毒感
染途径方法则以安全性高见长。如何建立兼备二者优点、符合临床要求的基因导入
法?一个方向是,更好地去除病毒上不需要的部分,以提高病毒感染途径的安全性
,同时无损于其高效性。另一种可能是,向脂类-DNA复合体或配体-多聚赖氨酸-
DNA复合体中加入其它因子,包括来自病毒的因子,以提高转导效能,同时无损于其
安全性。各种具体方法则根据治疗靶组织、治疗基因大小和数量、时效要求等的不
同而各有不同的用途。临床应用的进展有赖于基础研究的深入。
